高钙含量肽钙螯合物的制备与表征分析
Preparation and characterization analysis of peptide -calcium chelation with high calcium content

Abstract
[Objective]The calcium chelated peptides were prepared to improve the bioavailability of calcium.[Methods] The present study was designed to prepare and analyze the peptide-calcium chelates with high calcium content via the reaction between sheep bone collagen peptides(SBCP)and CaCl2.The optimal reaction conditions were determined by the overall consideration of chelating ratio,chelate yield rate and the calcium content in the chelate.[Results]The opti-mal condition was determined as SBCP at 90 mg·mL -1,the ratio of peptide to calcium at 2:1,pH 8.8,40oC,and re-action time at 30min.The average calcium content of the prepared chelate(SBCP -Ca)reached 9.81%,which was significantly higher than that at the highest chelating ratio(1.26%).After the chelating reaction,the loose flake mi-crostructure of SBCP turned into tight particles;m/z of the dominant fraction was increased from 1 230.57(SBCP)to 2 278.69(SBCP-Ca);the content of the proline was decreased from 10.96 g·100 g -1to 5.62 g·100 g -1,and chela-ting reaction was occurred at the point of O in -COOH and N in -NH2of the SBCP.[Conclusion]Study results indi-cated that all factors,including chelating rate,chelate yield rate,and the calcium content in the chelate(SBCP-Ca), should be taken into account when peptide-calcium chelates with high calcium content was prepared,and new com-pounds were formed during chelating reaction.关键词
