Effect of long chain non-coding RNA LINC01128 on proliferation and invasion of renal cell carcinoma and its mechanism

Abstract
Objective To observe the expression of long chain noncoding RNA LINC01128 in human renal cell carcinoma cell line and explore its effect on the proliferation and invasion of renal cell carcinoma and the mechanism.Methods The qPCR was used to detect the expression of LINC01128 in renal carcinoma cell lines A498,ACHN,786-O,OS-RC-2 and human proximal tubular epithelial cells HK-2.A498 cells in logarithmic growth phase were divided into control group and experimental group.Negative control plasmid(control group) and LINC01128 high-expressed plasmid (experimental group) were transiently transfected with Lipofectamine 2000,respectively.The expression of IncRNA LINC01128 and PTEN mRNA in A498 cells was detected by qPCR.The protein expression of PTEN,CDK4,Cyclin D1,E-cadherin and N-cadherin in A498 cells was detected by Western blot.Flow cytometry was used to determine the cell cycle of A498 cells.Clone formation assay and CCK-8 assay were used to detect the cell proliferation ability of A498 cells.Transwell assay was used to detect the cell invasion ability of A498 ceils.Results The expression of LINC01128 in renal cancer cell lines A498,ACHN,786-O,OS-RC-2 and human proximal tubular epithelial cells HK-2 was 0.18 ± 0.08,0.60 ± 0.10,0.44 ± 0.07,0.55 ± 0.13 and 1.02 ± 0.24,respectively.The expression of LINC01128 in renal cancer cell lines was higher than that of renal tubular epithelial cells HK-2(P ＜ 0.05).The expression levels of LINC01128 in control group and experimental group were respectively1.09 ± 0.57 and 72.27 ± 13.78 (t =10.320,P ＜ 0.001),and the expression of PTEN mRNA was respectively 1.03 ± 0.29 and 4.10± 1.15 (t =5.179,P =0.002).Compared with control group,the expression of PTEN and E-cadherin proteins increased in A498 cells in experimental group,while the expression of CDK4,Cyclin D1 and N-cadherin proteins decreased.Compared with control group,the cell cycle of A498 was inhibited in experimental group,while the proportion of cells in G0/G1 phase was increased(P ＜0.01).Compared with control group,the cell proliferation and invasive ability were decreased in experimental group (P ＜ 0.01).Conclusion LINC01128 can inhibit the proliferation and invasion ability of A498 cells by up-regulating the expression of PTEN gene,thereby inhibit the occurrence and development of renal cell carcinoma.关键词
