Abstract

Objective:Long-term obesity can cause male hypogonadism and low reproductive function.There is still much controversy over the effect of exercise on male reproductive function.The effect of exercise on the reproductive function of male rats with different body fat was studied by aerobic exercise intervention,and the effect on the expression of TNF-al-pha and Caspase-3 mRNA in the testicular tissue was explored from gene level,and the experimental basis was provided to reveal the mechanism of cell apoptosis.Methods:24 SD male rats were divided into two groups at random,namely normal diet group(common feed) and high fat diet group(high fat feed) for 12 weeks.After successful modeling of obese rats in high fat diet group,the two groups of rats were divided into 4 groups according to the weight stratification sampling:nor-mal control group(NC,6),normal exercise group(NE,6),obesity group(OC,6),obesity exercise group(OE,6).Two exercise groups were trained for 8 weeks of treadmill training,and the physical morphological indexes of the rats and the gene transcriptional level of the related factors were examined by measurement and RT-PCR.Results:1.Compared with NC group,the body weight and lipid ratio of OC group were significantly increased(P<0.01).The testicular coefficient, sperm count and sperm motility were significantly decreased(P<0.01).2.Compared with OC group,the body weight and lipid ratio of OE group were significantly decreased(P<0.01).The testicular coefficient,sperm count and sperm motility were significantly increased(P<0.01;P<0.01;P<0.05).3.Compared with the rats in NC group,the transcriptional lev-els of TNF-α,Fas and Caspase-3 in rats in the NE group were significantly decreased(P<0.05).The transcriptional lev-els of TNF-α,Fas and Caspase-3 in the testicular tissues of obese rats were significantly increased(P<0.05;P < 0.01;P< 0.01).The transcriptional levels of TNF-α,Fas and Caspase-3 in rats of the OE group were significantly lower than those of the OC group rats(P<0.01;P < 0.05;P < 0.01).Compared with NE group,the transcriptional level of TNF-α,Fas and Caspase-3 in rats of the OE group increased significantly (P<0.01;P < 0.01;P < 0.05).Conclusion: By raising the expression of the testicular TNF-αmRNA,the diet obesity activated the signal transduction of the death receptor pathway,which inhibited the development of the testicles and negatively impacted the male reproductive function.Long-term aerobic exercise can reduce TNF-α mRNA expression,inhibition of death receptor pathways of signal transduction, improve the negative impact of male obese rats on the testicular function,improve the reproductive function of normal male rats.
