摘要

Abstract

In this study,the expression of IKKγmRNA and proteins in the reproductive-and-respiratory syndrome-virus (PRRSV)-infected Marc-145 cell disease model was analyzed and the relationship between PRRSV and IKKγ in the NF-κB signaling pathway was revealed,which provided a theoretical foundation for research on the relationship between the pathway and viral infection.We used the indirect immunofluorescent assay (IFA) to determine if the model had been established successfully,and used the MT method to detect the LY294002 inhibition rate of the model cells.The model cells were divided into four groups:the control group,LY294002 group,PRRSV group and virus+LY294002 group.We also used qPCR and Western blot to detect the changes in expression of IKKγ and proteins in the different treatment groups so as to understand the molecular immune mechanism of IKKγin the anti-PRRSV process.The results of the qPCR showed that,compared with the control group,the IKKγ mRNA expression of the PRRSV group increased (P<0.05),but that of the LY294002 group (P<0.01) and that of the LY294002+PRRSV group both decreased (P<0.01).The results of Western blot showed that the expression of the IKKγprotein in the PRRSV group increased,but that of the LY294002 group and that of the LY294002+PRRSV group both decreased.The present study indicated that the LY294002 inhibitor had effect on the transcription and replication of PRRSV and that IKKγfunctioned in molecular immunization after PRRSV infection.
