Abstract

The primers were designed according to the Arabidopsis AtSPT cDNA sequence contained in GenBank,with the method of Homology-based cloning,the full-length cDNA of the CbSPT gene was amplified by RT-PCR in Capsella bursa-pastoris.Bioinformatics analysis showed that:the capsella bursa-pastoris CbSPT gene cDNA sequence 1191 bp, with 1047 bp open reading frames,coding 348 amino acids,the homology of nucleic acid sequence and amino acid se-quence of the AtSPT gene of Arabidopsis was 98%and 93%respectively,which could be judged as the CbSPT gene cDNA sequence(CbSPT).Based on Ti plasmid pWM101,CbSPT gene plant expression vector pWM101 -35S::CbSPT regulated by CaMV35S promoter was constructed.Arabidopsis thaliana was transformed by the method of agrobacterium,Arabidopsis plants transformed with CbSPT gene were obtained.There were some changes in the shape and size of the transgenic Arabi-dopsis carpel pods,which means that the expression of the CbSPT gene in Arabidopsis thaliana had a certain effect on the shape and size of Arabidopsis thaliana.But,it did not change the Arabidopsis into the shape of Capsella bursa-pastoris.
