摘要

Abstract

Objective:To optimize the extraction process and purification process of the total flavonoids from Rosa multiflora.Methods:The best combination of ethanol concentration,the ratio of material to liquid and extraction time was de-termined by L9(34)orthogonal experiment.The best model of the macroporous adsorption resin for purification was screened out with the method of static adsorption-desorb and dynamic adsorption-desorb,and also the best purifing conditions were made sure by investigating the single factor.Results:Under the UV 508nm detection wavelength,the best extractive combination was to reflux extract total flavonoids with 50% ethanol solution,the ratio of material to liquid 1:10 under 90 ℃,90 min.In 14 types of macroporous resin,HPD-400 was the best one in adsorption-desorb capacity.The best sample concentration was 1.5 times diluted with mother liquid,sample volume was 35 mL and the flow rate was 3 s/drop.The water volume for purifica-tion was 40 mL and the flow rate was 1 s/drop;50% ethanol solution for elution was 32 mL and the flow rate was 2 s/drop. Conclusion:Under the optimal conditions of extraction and purification,the content of the total flavonoid can reach above 50.00%.
