Abstract
Objective To investigate the effect and its mechanism of momordoside G on the proliferation,migration and apoptosis of human lung cancer A549 cells.Methods Different concentrations of momordoside G were applied to the invention on human lung cancer A549 cells as well as the A549 cells overexpressing CCAAT/enhancer binding protein β(C/EBPβ) and transforming growth factor β1(TGF-β1).EdU staining and methyl thiazolyl tetrazolium(MTT) assay were used to detect the proliferation and activity of human lung cancer A549 cells,Transwell assay to detect the migration of human lung cancer A549 cells,flow cytometry to detect the apoptosis of human lung cancer A549 cells,and Western blot and realtime quantitative PCR to determine the expression levels of C/EBPβ,TGF-β1and P53 as well as the activity of cysteinyl aspartate specific proteinase 3(Caspase3) of A549 cells.Results In the momordoside G intervention groups,the proliferation rate,viability and migration capacity of A549 cells were lower than those in the control group(all P<0.05),the apoptosis rate was higher than that in the control group,presenting a dose-independent relation(P<0.05),and the expression levels of matrix metalloproteinase (MMP)2 and MMP9 mRNA and protein in the momordoside G intervention groups were lower than those in the control group,presenting a dose-independent relation(P<0.05);the expression level of Caspase3 protein of human lung cancer A549 cells was higher than that in the control group(P<0.05),the expression levels of C/EBPβ and TGF-β1mRNA and protein were lower than those in the control group, and the levels of P53 mRNA and protein were higher than those in the control group(all P <0.05). After intervention with different concentrations of momordoside G,for human lung cancer A549 cells,the expression levels of C/EBPβ and TGF-β1in the pCDH-C/EBPβ and pCDH-TGF-β1groups were lower than those in the pCDH group,presenting a dose-independent relation(P<0.05).Conclusion Momordoside G can inhibit the proliferation and migration of human lung cancer A549 cells and induce cell apoptosis probably by dowregulating the expressions of C/EBPβ and TGF-β1and upregulating the expressions of Caspase3 and P53.关键词
