Abstract
Objective To evaluate the role of Janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) signaling pathway in the attenuation ofmyocardial ischemia-reperfusion (I/R) injury by oxycodone pretreatment in rats.Methods Forty-eight male SD rats,weighing 250-300 g,were randomly allocated into 4 groups (each of 12):sham operation (group S);group I/R;oxycodone pretreatment group (group OP);AG490 group (group OA).Taking the left anterior descending coronary artery for ligation of 30 min,and reperfusion of 120 min,to prepare for the myocardial ischemia-reperfusion injury model.Continuous recording of Ⅱ lead electrocardiogram (ecg).At 5 min before ligation via the internal jugular vein injection of oxycodone 0.5 mg/kg and at 30 min before oxycodone injection,AG490 3 mg/kg was injected intraperitoneally in group OPA.Blood samples were taken from the heart at 120 min of reperfusion for measurement of serum creatine kinase isoenzyme (CK-MB)and troponin (cTnI) activities.Myocardial infarct size was then measured by TTC staining method.The left ventricular myocardial tissues were obtained for histopathologic examination and for determination of the expression of Bax and Bcl-2 in myocardial tissues (immunohistochemically).Results Compared with group S,the serum CK-MB and cTnI concentrations were significantly increased,myocardial infarction area increased,and the expression of Bax and Bcl-2 in myocardial tissues was up-regulated;the ratio of Bcl-2 /Bax was reduced in I/R group,OP group and OA group.Compared with group I/R,the serum CK-MB and cTnI concentrations decreased,myocardial infarction area decreased,and the expression of Bax was down-regulated;Bcl-2 was up-regulated and the ratio of Bcl-2 / Bax was increased in myocardial tissues in OP group and OA group.Compared with group OP,the serum CK-MB and cTnI concentrations were increased,myocardial infarction area increased,and the expression of Bcl-2 was down-regulated;Bax was up-regulated and the ratio of Bcl-2 /Bax was reduced in OA group.Conclusion JAK2/STAT3 signaling pathway is involved in the attenuation of myocardial I/R injury by oxycodone pretreatment in rats.关键词
