摘要

Abstract

Objective To explore the effects of resveratrol in combination with curcumin on apoptosis of liver cancer cells. Methods Liver cancer cell line SMMC-7721 was used and divided into 4 groups, including group A (control group), group B (resveratrol group), group C (cur-cumin group) and group D (resveratrol+curcumin group) . CCK-8 (cell counting kit-8) method was used to detect the proliferative rates in different groups; colorimetry was employed to detect the activities of caspase-3 , caspase-8 and caspase-9; Western blotting was utilized to detect the expres-sion of Bax, Bcl-2, PARP ( poly-ADP-ribose polymerase) and Cleaved PARP. Results CCK-8 results showed that the proliferative rate was lowest in group D ( P<0. 05 ) . Colorimetric results exhibited that the activity of caspase-3 , caspase-8 and caspase-9 was highest in group D ( P <0. 05) . Western blotting revealed that Bax expression in group D was higher, and Bcl-2 expression was lower in group D than in group A, B and C ( P<0. 05 for all) . PARP expression was lower in group D and cleaved PARP was higher in group D than in group A, B and C (P<0. 05) . Con-clusion Resveratrol and curcumin can induce the apoptosis in SMMC-7721 cells, and the combi-nation of both can enhance the effects, possibly by increasing the caspase activity and Bax expres-sion, and decreasing Bcl-2 expression and the cleavage of PARP.
