摘要

Abstract

Objective To investigate effects of LncRNA-GAS5 knockdown or overexpression on human mononu-clear macrophage apoptosis. Methods Knockdown or overexpressed LncRNA-GAS5 of human mononuclear macrophage cell lines THP-1 were established by using siRNA or LncRNA-GAS5 expression plasmids. Conditions of cell apoptosis were detected using Annexin V-PE/ 7-AAD kits and flow cytometry methods. Western blot was used to detect expression of associated apoptotic protein. Results Apoptotic percentage of THP-1 cells in low-expressed LncRNA-GAS5 group was significantly higher than that in normal-expressed LncRNA-GAS5 group. There was no significant difference in apoptotic percentage of THP-1 cells between over-expressed and normal-expressed LncRNA-GAS5 groups; while under ox-LDL (75 μg/ ml, 24 h) stimulation, apoptotic percentage of THP-1 cells in low-expressed LncRNA-GAS5 group was signifi-cantly increased compared with that in normal-expressed LncRNA-GAS5 group. Apoptotic percentage of THP-1 cells in o-ver-expressed LncRNA-GAS5 group was significantly decreased compared with that in normal-expressed LncRNA-GAS5 group. Analysis of apoptotic protein showed that under ox-LDL (75 μg/ ml, 24 h) stimulation, overexpression of Ln-cRNA-GAS5 significantly decreased protein expressions of caspase-9, caspase-7 and caspase-3, while knockdown of Ln-cRNA-GAS5 significantly promoted the above protein expressions. Conclusion LncRNA-GAS5 may obviously inhibit hu-man mononuclear macrophage apoptosis.
