Abstract
The aim of this study was to investigate effects of COL6A1 down-regulation by RNA interference ( RNAi) on proliferation and invasion of renal carcinoma cell line 786-0. SiRNAs targeting COL6A1 gene were designed, and transi-ently transfected into renal carcinoma cell line 786-0. The transcription level of COL6A1 was detected by RT-PCR. Activi-ties of COL6A1 protein were measured by Western blotting. The cell proliferation was detected by MTT at day 1, day 2, and day 3 after transfection. The cell' s invasion ability was performed on Matrigel transwell assay. The results showed that the levels of COL6A1 mRNA and protein was significantly reduced after RNAi( P<0.05) . MTT assay showed that the prolif-eration ability of 786-0 cell was decreased due to RNA interference( P<0.05) . Transwell assay showed that invasion ability of 786-0 cell was reduced obviously due to COL6A1 RNAi( P<0.05) . The ability of proliferation and invasion in 786-0 cells can be inhibited by RNAi-targeting COL6A1.关键词
