荔枝核总黄酮对TGF-β1诱导人肝星状细胞凋亡的影响及机制
Effects of total flavonoids of semen litchi on TGF-β1-induced apoptosis of human hepatic stellate cells

Abstract
Objective To investigate the effects of total flavonoids of semen litchi (TFL) on the apoptosis of human hepatic stellate cells LX2(HSC-LX2) induced by transforming growth factor β1 (TGF-β1),and to explore the related molecular mechanisms.Methods The cultured HSC-LX2 cells were randomly divided into the normal group,TGF-β1 group,and the different concentrations of TFL groups (150,300,and 600 mg/L TFL).Cells in the TGF-β1 group and different concentrations of TFL groups (150,300,and 600 mg/L TFL) were cultured in the culture dish with DMEM (10% FBS) and TGF-β1 (5 μg/L);after 24-hour culture,different concentrations of TFL groups (150,300,and 600 mg/L TFL) were added with different concentrations of TFL (150,300,and 600 mg/L TFL);after 48 h,the related indicators were measured.The morphological changes of cells were observed by Hoechst33258 staining;the cell cycle was detected by flow cytometry;FITC Annexin V/PI double staining was used to detect the apoptosis;the expression levels of TLR4,NF-κB,and IL-1R Ⅰ proteins were detected by Western blotting.Results The nuclear morphology of cells in the normal group and the TGF-β1 group was complete,the proliferation was active in the TGF-β1 group,and the morphological changes of apoptosis including pyknosis,lysis and apoptotic bodies of nucleus were observed in the different concentrations of TFL groups (150,300,and 600 mg/L TFL).Flow cytometry showed that with the increasing TFL concentrations,the cells in the G0/G1 phase increased in the TFL groups (P < 0.05),decreased in the S phase (P < 0.05);with the increasing TFL concentrations,TFL promoted the apoptosis of HSC-LX2,and tended to promote late apoptotic.In the different concentrations of TFL groups (150,300,and 600 mg/L TFL),TLR4,NF-κB,and IL-1R Ⅰ protein expression was lower than that of the TGF-β1 group (P <0.05);with the increase of TFL concentrations,the expression of TLR4,NF-κB,and IL-1R Ⅰprotein in HSC-LX2 decreased gradually (all P < 0.05).Conclusion TFL can promote the TGF-31-induced apoptosis of HSC-LX2 cells,especially for the late apoptosis,by reducing the expression of TLR4,NF-κB,and IL-1R Ⅰ.关键词
