Screening microRNA expression profile in patients with acute rejection after renal transplantation

Abstract
Objective To investigate the expression changes of serum microRNA (miRNA) in patients with acute rejection (AR) after renal transplantation.Methods The elbow venous blood from 26 cases (13 AR patients and 13 non-AR patients) of patients after renal transplantation (research group) and 10 cases of healthy volunteers (control group) was collected.Serum samples from 3 AR patients and 3 non-AR patients were randomly selected,and the microarray analysis was used to identify the miRNAs profile.According to the results of miRNA marker screening,the expression of miR-142-5p,miR-144-5p,and miR-130a-3p in the remaining 10 AR patients,10 non-AR patients and 10 healthy controls were detected by PCR.Results The analysis of microarray showed that 96 miRNAs had differential expression in the AR patients and non-AR patients,including 13 (13.5％) up-regulated miRNAs and 7 (7.3％) down-regulated miRNAs.The expression levels of miR-142-5p,miR-144-5p,and miR-130a-3p in the AR patients were significantly higher than those of the non-AR patients and healthy volunteers.Conclusion The levels of miR-142-5p,miR-144-5p and miR-130a-3p in AR patients after renal transplantation are highly expressed.关键词
