Relationships of NOD1 rs2075819A>G and rs2075820G>A genetic variants with risk of lung cancer

Abstract
Objective To explore the relationships of rs2075819A > G and rs2075820G > A genetic variants in nucleotide-binding oligomerization domain-containing protein 1 (NOD1) gene with the risk of lung cancer.Methods A total of 706 non-small-cell lung cancer (NSCLC) patients and 706 healthy controls were included.The genotypes of NOD1 rs2075819A > G and rs2075820G > A variants were determined by polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP).The OR and 95% CI were estimated by Logistic regression to evaluate the effects of genotypes of rs2075819 and rs2075820 on the risk of lung cancer.Results Compared with the NOD1 rs2075819AA genotype carriers,AG genotype carriers had a significant lower risk of NSCLC (OR =0.79,95 % CI =0.63-0.99,P =0.044).When stratified by age for rs2075819,AG genotype carriers with the age ≤60 had a decreased risk (OR =0.69,95% CI =0.51-0.94,P =0.018).When stratified by smoking status for rs2075819,as for non-smokers,AG genotype carriers had a decreased risk (OR =0.74,95% CI =0.56-0.98,P =0.037).There was no significant difference in NOD1 rs2075820 genotype distribution between patients and controls,as well as in age and smoking stratification (all P > 0.05).Conclusions The mutation of rs2075819A > G in the NOD1 gene may affect the risk of lung cancer,and carriers with the rs2075819AG genotype have a lower risk of lung cancer.s2075820G > A mutation does not affect the risk of lung cancer.关键词
