摘要

Abstract

The asymmetric synthesis of chiral amines and unnatural amino acids using ω-transaminase as catalyst is one of the hot topics in bioprocess engineering. Because of its excellent stereo- and regio-selectivity,ω-transaminase has been widely used in the fields of medicine, pesticide and chemical industry.This review describes the basic structural characteristics of ω-transaminase and summarizes the recent research advances in the high-throughput screening assay development and protein engineering ofω-transaminase. The applications of ω-transaminase in asymmetric synthesis of chiral amines and the cascade-involved strategies for increasing chiral amine production are then discussed ( sitagliptin synthesis as an example was emphasized).Finally,the synthesis of unnatural amino acids by ω-transaminase was briefly introduced.
