戊二醛交联的传感器检测布鲁氏菌抗体的研究
Developing glutaraldehyde cross-linking based immunosensors for Brucella melitensis antibody detection
摘要
Abstract
[Objective] A novel method was established to detect minimum low concentrations of Brucella melitensis an-tibody for early diagnosis of brucellosis .[Methods]Both original and ultrasonic broken Brucella melitensis antigens were crosslinked and entrapped on the pretreated screen-printed gold electrode surface by glutaraldehyde .Then the im-mune responses of antigens were characterized by cyclic voltammetry ,different wave voltammetry ,and square pulse voltammetry ,and a label-free amperometic immunosensor was constructed .[Results] A linear function between peak current and the logarithm concentration of Brucella melitensis antibody was found in the range of 1 × 10 - 5 ~ 1 × 10 IU·mL - 1 .The immunosensor system tested by square wave voltammetry exhibited the highest sensitivity ,and the lowest detection limit at 1.2065 × 10 - 9 IU·mL - 1 .[Conclusion] The results showed that the immunosensor pretreated with glutaraldehyde cross-linking could effectively detect Brucella melitensis antibody at low concentration .关键词
