Effects of Early Weaning on Glycogen Concent and Expression of Hepatic Genes Associated with Gluconeogenesis and Glycolysis in Piglets

Abstract
The objective of this study was to elucidate the effects of early weaning on glycometabolism of piglets.Sixteen 18-day-old piglets were selected from 4 litters with each litter selecting 4 piglets.After weighted,four piglets were euthanized directly and serum,liver and longissimus muscle samples were collected,while the other 12 piglets were segregated from the sows as the weaned group.The rest of the piglets in the same litter continued being breastfed as suckling group.Four piglets were weighted on day 1,3 and 7 postweaning in weaned group and age-match suckling group,respectively,and then used to measure the contents of glucose and lactate in serum,the content of glycogen in liver and longissimus muscle,the activity of pyruvate kinase and the relative expression level of genes associated glycometabolism in liver.The results showed that the average daily gain of piglets in weaned group was significantly decreased compared with suckling group at early weaning stage (1 to 7 days postweaning) (P<0.05).Compared with suckling piglets,serum glucose content of weaned piglets was decreased by 8.33% (P>0.05),17.81% (P<0.05) and 20.99% (P<0.05) on day 1,3 and 7 postweaning,respectively;whereas serum lactate content of weaned piglet was increased by 16.83% (P<0.05),22.75% (P<0.05) and 12.06% (P<0.05),respectively.Additionally,the liver glycogen content of weaned piglets was decreased by 24.94% (P<0.05),48.99% (P<0.05) and 36.51% (P<0.05),respectively;and muscle glycogen content of weaned piglets was decreased by 44.49% (P<0.05),39.68% (P<0.05) and 25.52% (P<0.05),respectively.Compared with suckling piglets,the activity of pyruvate kinase,a key glycolytic enzyme,and its gene relative expression level in liver of weaned piglets were significantly decreased on day 3 and 7 postweaning (P<0.05),while the relative expression levels of glycogen related enzymes,such as mitochondrial phosphoenolpyruvate carboxykinase (day 1 postweaning),cytoplasmic phosphoenolpyruvate carboxykinase (day 1,3 and 7 postweaning) and glucose-6 phosphatase (day 1 and 3 postweaning) in liver of weaned piglets were significantly increased (P<0.05).In conclusion,the contents of serum glucose,liver glycogen and muscle glycogen are decreased in piglets by early weaning,and the gluconeogenesis is activated and the glycolysis is suppressed in liver of early weaned piglets to ensure the balance of glycometabolism in body.关键词
