急性痛风性关节炎大鼠P2Y6的表达水平及意义
Expression level of P2Y6 and its significance in rats with acute gouty arthritis

Abstract
Objective To explore the expression level of P2Y6 and its significance in rats with acute gouty arthritis. Methods Thirty male SD rats were randomly divided into control group,model group and colchicine group,with 10 rats in each group. The colchicine group was given colchicine by gavage,and the control group and the model group were given the equivalent amount of normal saline by gavage. On the third day after intragastric administration,monosodium urate(20 mg/ml) was injected into right ankle of the rats in the model group and colchicine group,50 μl per rat,while the injection of the equivalent amount of normal saline was performed in the control groups. Then to stop intragastric administration on the fifth day. The swollen joint count index,pathological changes of joint cartilage,and expressions of P2Y6 mRNA and protein in ankle tissues were compared among the three groups after modeling. Results The swollen joint count indices in the model group were higher than those in the other two groups at 4 h,12 h,24 h and 48 h after molding(P<0. 05),and at 12 h and 24 h after modeling,the swollen joint count indices in the colchicine group were higher than those in the control group but lower than those in the modeling group(P<0. 05). Microscopic findings revealed that obvious inflammatory cell infiltration and active proliferation of cartilage cells were observed in ankle tissues of the model group,but the features above were milder in the colchicine group. The expression levels of P2Y6 mRNA and protein increased in turn in the normal group,the colchicine group and the model group(P<0. 05). Conclusion Elevated P2Y6 level is seen in the rats with acute gouty arthritis. P2Y6 might play an important role in the incidence of acute gouty arthritis, and it might be a new therapeutic target.关键词
