摘要

Abstract

Aimed to reveal the cold tolerance and adaptive mechanism in leaf of Distylium buxifolium during natural temperature changes in Qingdao,leaves of D.buxifolium were used as materials.The leaf tissue structure,cell cytoplasm membrane permeation and physiological changes of D.buxifolium were investigated by routine paraffin section and determination of relative conductivity and relevant physiological indexes during natural temperature changes in Qingdao.Experimental results showed that the leaf tissue structure and physiological metabolism activities were affected in different degrees with the temperature changed.From October 2015 to April 2016,the temperature reduced at first and then increased.Different indexes of D.buxifolium leaf including relative conductivity,SOD activity,MDA content,soluble protein content and proline content were analyzed,which tend to increase first and then decrease with the temperature.The indexes of leaf thickness and ratio of cell length and width in palisade displayed significantly higher in January 2016 than that in October 2015 and April 2016,while relative volume percentage ratio and cell volume of palisade tissue showed an opposite trend with lower in April 2016.The above results indicate that improved ability of cold tolerance in D.buxifolium relies on the changes of leaf tissue structure and physiological metabolism activities.
