Effect of Butyric Acid On Growth and Apoptosis of Rumen Epithelial Cells

Abstract
The aim of this study was to investigate the effect of butyric acid on the growth and apoptosis of rumen epithelial cells in vitro. Rumen epithelial cells were treated with different concentrations of butyrate acid (0,20,40,60,80,100,120,140 mmol/L) and different treatment time (4,6,8,10 h).After that,the viability of epithelial cells in each group was detected by MTT assay. Then the suitable treatment group was used to detect cell apoptosis by TUNEL assay,DNA ladder assay and flow cytometry. The results showed that:with the increase of butyric acid concentration,the relative cell viability of rumen epithelial cells increased firstly,and then decreased.Additive of 20 mmol/L butyric acid promoted the growth of rumen epithelial cells, while 40-140 mmol/L butyrate decreased the viability of ruminal epithelial cells.Apoptosis detection revealed that apoptosis of rumen epithelial cells induced by the additive of 120 mmol/L butyric acid cultured for 6 h. The total apoptosis index of epithelial cells in 120 mmol/L butyrate group was 28.4%,which was significantly higher than that in the control group (24.1%,P<0.05).In conclusion,low concentration of butyric acid promo?ted the growth rate of rumen epithelial cells,while high concentration of butyric acid induced the apoptosis of rumen epithelial cells.关键词
