Effect of advanced glycation end products on ovarian primordial follicles

Abstract
Objective:To investigate the effect of advanced glycation end products on the activation of primordial follicles.Methods:The ovaries of ICR mice with 4 day old were cultured in basic culture medium,200 μg/ml BSA medium and 200 μg/ml AGE-BSA medium respectively for 4 days,and then the ovary tissues were taken.HE staining was used to observe the composition ratio of ovarian follicles in different development stages.Western blot was used to detect the receptor of AGEs(RAGE)and the follicular activation markers (PCNA).The localization of PCNA was detected by immunohistochemistry.Results:The growth follicular composition ratio in AGE-BSA group was significantly higher than solvent control group (P < 0.001),with the increase of primordial follicles in ovarian medulla.The expression of PCNA and RAGE were significantly higher in AGE-BSA group(P<0.05).However,there were no significant differences in follicular composition ratio and the expression of PCNA and RAGE between the control group and the solvent control group(P>0.05).The immunohistochemical results showed that PCNA was expressed in ovarian follicle oocytes and granulosa cells at all the stages.Conclusions:Advanced glycation end products promote the activation of primordial follicles through AGEs-RAGE axis,and promote follicular pool depletion.关键词
