Abstract
Objective:To explore the molecular mechanism of arsenite-mediated regulation of embryonic p66Shc expression.Methods:After 5 μmol/L sodium arsenite treatment,the transcriptional level of p66Shc was detected by real-time quantitative PCR analysis and the expression of Sirt1 was detected by Western blot.The cells were transfected with Sirt1 or control siRNA.At 24 hours post-transfection,cells were exposed to 5 μmol/L sodium arsenite for another 24 hours,and proteins were extracted for Western blot analysis to detect the effect of sodium arsenite on the expression of Sirt1 and p66Shc proteins.Results:The expression of p66Shc on transcriptional level was significantly increased in sodium arsenite treated-group compared with the control group(P<0.001).Expression of Sirt1 was significantly decreased in sodium arsenite treated-group compared with the control group(P<0.001).Transfection of Sirt1 siRNA could repress the expression of Sirt1.After sodium arsenite treatment,Sirt1 expression was significantly decreased in Sirt1 siRNA transfection group compared with control siRNA transfection group (P<0.001),while p66Shc expression was significantly increased (P<0.001).Conclusions:Sodium arsenite may up-regulate p66Shc expression by inhibiting the expression of Sirt1.关键词
