原代大鼠脑微血管内皮细胞的培养及鉴定
Primary culture and identification of microvascular endothelial cells of rat brains

Abstract
Objective:To establish a method that can successfully separate and cultivate microvascular endothelial cells of rat brains in vitro.Methods:The microvascular segments of the rat brain were obtained from 7 to 10 d SD rats by homogenation,bovine serum albumin gradient centrifugation and digestion with collagenase type Ⅱ and then they were cultured in carbon dioxide incubator.The cultured cells were identified by cell morphology observation and immunohistochemical detection of Ⅷ factor-related antigen.Results:Under the inverted microscope,the cells in vitro cultured for 24 h to 48 h,emerged from the microvascular section of the wall.The cells were fusiform-shaped or polygonal-shaped,and proliferated in a clustered monolayer.The typical "paving pebbles" sample was observed after the cells became dense.The correlation antigen of Ⅷ factor was positive,the cytoplasm was brown,and the positive cells accounted for more than 99%.Conclusion:The method can successfully separate and cultivate microvascular endothelial cells of rat brains in vitro.关键词
