骨髓间充质干细胞与硫酸软骨素酶ABC联合移植对大鼠脊髓损伤的修复作用及其机制
Transplantation of ChABC combined with BMSCs helps repair spinal cord injury in rats

Abstract
Objective To explore the role and related mechanism of combined transplantation of chondroitinase ABC (ChABC) and bone marrow stromal cells (BMSCs) during the repair process of rats with spinal cord injury.Methods Eighty SD rats were randomly divided into four groups (A,B,C,and D) with 20 rats in each group.On the sixth day after injury,the rats in group A were transplanted with 20 μL of 2.0 × 106/μ L original BMSCs suspension and 1 U/mL ChABC,while the same dose of BMSCs,ChABC,and PBS were transplanted into group B,C,and D respectively.The BBB score,spinal cord injury area,somatosensory evoked potential (SEP),motor evoked potential (MEP) latency,and the percentage of cells with positive expression of glial fibrillary acidic protein (GFAP),growth-associated protein 43 (GAP 43),and brain-derived neurotrophic factor (BDNF) were compared among different groups.Results Compared with group D,BBB scores of the other three groups were lower on day 1 after transplantation,and became higher on the 14th and 28th day after transplantation.Compared with group A,BBB scores of group B and C were lower (both P < 0.05).Compared with the same group before treatment,the latency of SEP and MEP in the groups A,B and C was short;compared with group A,the latency of SEP in the group B was longer while shorter in the groups C and D;compared with group A,the latency in the other three groups were shorter (all P < 0.05).The injury area was the smallest in the group A as compared with that of the other three groups (all P < 0.05).Compared with group D,the percentage of GFAP positive cells was lower while the percentage of GAP-43 and BNDF positive cells was higher in the other three groups.Compared with group A,the percent-age of GFAP positive cells was higher while the percentage of GAP-43 and BNDF positive cells was lower in the other three groups (all P < 0.05).Conclusion Transplantation of ChABC combined with BMSCs can help repair the spinal cord injury in rats,and the mechanism may be related to the down-regulation of the GFAP,GAP-43 and the up-regulation of BDNF.关键词
