猪胚胎期小肠组织MKRN3基因启动子区CpG岛的甲基化模式分析
The Methylation Model of Porcine MKRN3 Promoter CpGisland in Embryo'S Intestine

Abstract
To identify the methylation model of porcine MKRN3 promoter CpG island in embryo's intestine,we utilized the 65 days and 100 days embryos from the reciprocal crosses of Yorkshire and Meishan pig.Firstly,CpG island in the promoter region of MKRN3 was predicted by utilizing bio-informatics method,and a pair of primers was designed based on this predicted CpG island.Bisulfite sequencing was utilized to detect the methylation level in embryos from Yorkshire-Meishan reciprocal crosses at different developmental stages.Results showed that,in the Yorkshire×Meishan 65 days embryo's intestine,the CpG island of porcine MKRN3 showed high level of methylation (70.8%),but in Meishan×Yorkshire 65 days embryo's intestine,it showed low level of methylateon (8.4%),which indicated a exactly opposite methylation model.In Yorkshire×Meishan 100 days embryo's intestine,the CpG island of target gene showed low level of methylation (16.4%),while in Meishan×Yorkshire 100 days embryo's intestine,the average methylation level was 37.2%.Results above indicated that the methylation model of porcine MKRN3 promoter CpG island would change regularly according to the change of reciprocal crosses and developmental stages.Thus,we infer that the DNA methylation of MKRN3 has parental preference,and demethylation might take place in the paternal or maternal allele,during the stage of 65 day and 100 day.Our study provided a foundation for the identification of how methylation regulate the gene expression and imprinting status.关键词
