摘要

Abstract

By comparing the maturation of oocytes in vivo and in Vitro in synthesizing glutathione related genes,and trying to use Sulforaphane to increase GSH synthesis in oocytes.Three weeks mouse were used in this experiments,and found that in the natural mature oocytes Gclc,Gclm near MⅡ was higher,and the cumulus cells near MⅠ was higher.The expression of Gclc and Gclm in vitro maturation oocytes was significantly lower than that in vitro in both MⅠand MⅡ (P<0.05),while the expression of MⅠ was lower than that in vivo.There was no difference in Gss.After that,we attempted to add 1,5,10,20 μmol/L Sulforaphane to improve the content of GSH in oocytes.The results showed that adding 10 μmol/L SFN can significantly improve the mRNA level of GSH related genes (P<0.05),and eventually improved the GSH content of the oocytes in vitro in order to achieve the purpose of optimizing the system of in vitro maturation.
