摘要

Abstract

Objective To investigate the effect of focal cerebral ischemia-reperfusion injury on the expression of a-smooth muscle actin (α-SMA) in mouse brain smooth muscle cells. Methods The middle cerebral artery occlusion (MCAO model) was established by right cerebral embolism ischemia in the C57BL/6 mouse model. The expressions of a-SMA in the smooth muscle cells of the cerebral ischemia reperfusion mice cerebral ischemia 2 h reperfusion 24 h group (the experimental group) and the sham operation group and the blank control group were observed by qRT-PCR, Western blot and immunohistochemistry. Results TTC (2, 3, 5-triphenyltetrazolium chloride )staining showed that the cerebral infarct size of the experimental group was significantly higher than that of the sham operation group and the blank control group. The experimental group had obvious cerebral edema. Western blot and qRT-PCR showed that the expression of a-SMA in focal cerebral ischemia-reperfusion group was significantly higher than that in sham operation group and blank control group at 2 h after reperfusion for 24 h (P < 0. 05). Conclusion Focal cerebral ischemia-reperfusion injury can stimulate the brain smooth muscle cells expression of a-SMA. The expression of a-SMA in brain is significantly higher than that in sham operation group and blank control group at 2 h after reperfusion.
