Gli promote epithelial-mesenchymal transition in esophageal adenocarcinoma by PI3K/AKT pathway

Abstract
Objective To investigate the mechanism of Gli regulating the epithelial-mesenchymal transition ( EMT) of esophageal squamous cell carcinoma KYSE-30 cells. Methods The mRNA expression of Gli1,Gli2,Vimentin and N-cadherin were detected after treatment with GANT61 by real time fluorescence quantitative PCR method in e-sophageal squamous cell carcinoma KYSE-30. The protein expression of Gli1,Gli2,p-AKT,AKT,Vimentin and N-cadherin were observed by Western blot assay after treatment with GANT61. Transwell assay were performed to de-tect cell invasion ability. The Hedgehog pathway was stimulated by N-Shh, and the Hedgehog pathway was stimula-ted by GANT61 and N-Shh. Results Compared with DMSO, GANT61 could significantly inhibit the mRNA ex-pression ( all P<0. 01) in KYSE-30 cell lines. Western blot assay revealed that GANT61 can down-regulate the protein expression in KYSE-30 cell lines(all P<0. 001). Furthermore, GANT61 could dramatically inhibit inva-sion in KYSE-30 by Transwell assay ( P<0. 001) . N-Shh can significantly up-regulate corresponding mRNA and protein expression, and promote the invasion of tumor cells (P<0. 001),GANT61&N-Shh group was partially re-stored compared to GANT61 group. Conclusion Our data highlight the importance of Shh pathway for metastasis of esophageal squamous carcinoma, and it may be related to promote epithelial-mesenchymal transition through PI3K/AKT pathway. Gli could be a therapeutic target in eliminating esophageal squamous carcinoma.关键词
