摘要

Abstract

Objective To study the expression of NLRP3 in bladder cancer cell lines and its effects on proliferation, migration and invasion of bladder cancer cells. Methods 31 cases of bladder cancer tissue samples and 8 adjacent tissues were obtained from the patients who underwent surgical resection. The real-time fluorescence quantitative PCR( qPCR) and Western blot were used to assess the expression of NLRP3. The correlation between the clinical data and the expression of NLRP3 was analyzed. In vitro, bladder cancer cells ( T24, 5637 and BIU-87) were used to investigate the effects of NLRP3 on proliferation, migration and invasion of cancer cells. Results Compared with the adjacent tissues, the expression of NLRP3 mRNA and protein in bladder cancer tissues was significantly up-regulated. Statistical analysis showed that the expression of NLRP3 mRNA was significantly correlated with pathological stage ( pT) , lymph node status( pN) and WHO tumor grading ( all P < 0. 01) . The expression of NLRP3 in tumor cell lines was significantly higher than that in normal bladder epithelial cells ( P < 0. 01) . The expression of NLRP3 in 5637 and T24 cells was increased significantly( P < 0. 01) . Silencing of NLRP3 significantly inhibited cell proliferation, migration and invasion of 5637 and T24 cells in vitro. Conclusion The expression of NLRP3 is closely correlated to the malignant characteristics of bladder cancer, the proliferation, migration and invasion ability of bladder cancer, and it is expected to be a potential target for bladder cancer treatment.
