Abstract

Objective To investigate the influence of propofol on invasion , metastasis and epithelial -mesenchy-mal transition (EMT) of human hepatocellular carcinoma cells .Methods Hepatocellular carcinoma cells ( HCCLM3, QGY-7703, SMMC-7721) were stimulated with propofol in gradually increasing dosages (5 μg/mL, 10 μg/mL and 20 μg/mL).The effects of propofol on the migration and invasion of hepatocellular carcinoma cells were assessed by wound healing assay, trans-well matrix invasion assay and three -dimensional spheroid invasion assay .HCCLM3 cells were stimulated with propofol of 20 μg/mL.Western blot and immunofluorescence assay were used to detect the expression of EMT makers (E-cadherin and Vimentin ).Results With the increase of propofol concentration , wound healing assay showed that the migration ability of hepatocellular carcinoma cells significantly decreased ( P<0.05); trans-well inva-sion experiments and three -dimensional culture experiments showed that the invasive ability of hepatocellular carcinoma cells significantly decreased (P<0.05).The expression of E-cadherin was up-regulated and the expression of vimen-tin was down-regulated in HCCLM3 cells after propofol stimulation of 20 μg/mL (P<0.05).Conclusion In vitro, propofol can suppress the migration and invasion of HCC cells with concentration -dependent effect , which may be corre-lated to the inhibition of EMT.
