miR-135b对LZST1表达及肺癌细胞转移行为的影响
Effects of miR-135b on LZST1 Expression and Metastasis of Lung Cancer Cells

Abstract
Objective To investigate effects and its mechanism of miR-135b on leucine zipper tumor suppressor gene 1 (LZST1) expression and metastasis of lung cancer cells. Methods Expressions of miR-135b and LZST1 in lung cancer tissues and adjacent normal tissues were detected by quantitative polymerase chain reaction (qPCR). Expression of miR-135b in lung cancer A549 cells was up-regulated and down-regulated by transfecting miR-135b-mimic and miR-135b-inhibitor mimics. Western blotting was used to detect regulatory effect of miR-135b on LZST1. Scratch healing as-say and Transwell invasion assay were used to detect effects of miR-135b on migration and invasion of lung cancer cells. Immunohistochemistry method was used to detect expressions of LZTS1 protein in lung cancer tissues and adjacent normal tissues. Results Expression levels of miR-135b and LZST1 in lung cancer tissues were higher than those in adjacent normal tissues (P<0. 05). Expressions of LZST1 protein were positive in lung cancer tissues and negative in adjacent normal tissues. Compared with those in negative control group, expression of LZST1 protein in miR-135b-mimic group was up-regulated, while the expression in miR-135b-inhibitor group was down-regulated (P <0. 05). Compared with those in negative control group, migration and invasion abilities in miR-135b-mimic group were enhanced, while the abil-ities in group miR-135b-inhibitor were weakened (P<0. 05). Conclusion The miR-135b may affect migration and in-vasion abilities of lung cancer A549 cells by regulating expression of LZST1 in lung cancer cell.关键词
