Abstract

This paper was to study a nanoparticle preparation for swine ferritin sub-units.Firstly,a pair of specific primers for the ferritin heavy chain gene were designed,whose sequence was referred on the GenBank website.Total RNA was extracted from the porcine heart sample,and the gene segment for the H-chain subunit was amplified by RT-PCR.The verified gene segment was integrated into the PET32a vector,and this recombinant plasmid was similarly clarified by PCR amplification,dual-enzyme digestion,and sequencing.The plasmid was then transformed into E.coli BL21(DE3)and protein expression was induced by isopropyl-β-D-thiogalactoside(IPTG).The expressed recombinant protein was purified by Ni+ion affinity chromatography,and identified by SDS-PAGE and Western blot.Transmission electron microscopy(TEM)was employed to visualize the size of protein.The results showed that the ferritin H-chain subunit was successfully expressed by E.coli BL21,and particles with diameter range of 10nm and 13 nm were observed.It indicated that the nanoparticles of swine ferritin sub-units were successfully obtained assemblied.
