摘要

Abstract

In order to improve the tobacco quality by degrading the cellulose,several cellulose-degrading strains were isolated from the surface of tobacco leaves.The activity of cellulose was detected,and the strain with the highest cellulase-producing activity was identified by the 16 S rDNA gene sequence and physiological-biochemical tests.Orthogonal experiment was further designed to get the optimal fermentation conditions of cellulose degrading in the tobacco leaves.The results showed that:(1)strain XC-19-1 had the highest cellulase enzyme producing activity(6.6 U·mL-1)among all eleven isolated strains;(2)strain XC-19-1 was identified as Bacillus subtilis by the 16 S rDNA gene sequence and physiological-biochemical tests;(3)the orthogonal experiment result suggested that,by adding 3%culture medium(with strain XC-19-1)to the surface of tobacco leaf,the degrading rates of cellulose could reach 30.6%after 60 hours fermentation under the condition of 37 ℃ temperature and 75%humidity.The contents of total sugar and reducing sugar were both increased after fermentation,which indicated the improvement of tobacco quality.
