FGF4在肺癌组织中的表达及其对肺癌细胞增殖、转移的影响
Expression of FGF4 in lung cancer and its effect on proliferation and metastasis of lung cancer cells

Abstract
Objective To observe the expression of fibroblast growth factor 4 (FGF4) in the lung cancer tissues,and to discuss the effects of FGF4 on the proliferation and metastasis of lung cancer cells. Methods We selected 40 cases of lung cancer tissues and their corresponding paracancerous tissues,human lung fibroblast cell line HFL1,human low metastatic lung cancer cell line NL9980,and human high metastatic lung cancer cell line A549. The mRNA expression of FGF4 in the tissues and cells was detected by RT-PCR,and the protein expression of FGF4 in the cells was detected by Western blotting. NlL9980 cells were transfected with Lipo2000 to construct stable cell line NL9980-FGF4-OE with over-expression of FGF4. The negative controls were NL9980 cell line only transfected with pCDNA3.1,that was NL9980-NC. A549 cells were transfected with Lipo2000 to construct the stable cell line A549-FGF4-si with knockout of FGF4. The negative control was A549 cell line transfected with pCDNA3.1,namely A549-NC. Meanwhile,the proliferation ability of NL9980-NC, NL9980-FGF4-oe,A549-NC,and A549-FGF4-si cells was detected by MTT assay,and the cell migration ability was detected by Scratch test. Results The mRNA expression of FGF4 in the lung cancer tissues was higher than that of the adjacent tissues (P < 0.05). What's more,the mRNA and protein expression levels of FGF4 was in the following order: A549> NL9980 > HFL1 (all P < 0.05). The proliferation capacity of NL9980-FGF4-oe cells was higher than that of NL9980-NC,at the same time,the proliferation capacity of A549-FGF4-si cells was lower than that of A549-NC cells (both P <0.05). The migration distance of NL9980-FGF4-oe cells was greater than that of NL9980-NC cells,meanwhile,the migration distance of A549-FGF4-si cells was shorter than A549-NC (both P < 0.05). Conclusion FGF4 is highly expressed in the lung cancer tissues,and its over-expression can promote the proliferation and metastasis of lung cancer cells.关键词
