胚胎培养微滴内的温度变化研究
Temperature variation in embryo culture micro-drops

Abstract
Objective:To investigate the temperature changes in embryo culture micro-drops when the Petri dish was taken out from the incubator and put on the thermo plate,and explore the time required to return to the setting temperature in 4 kinds of incubators. Methods: Four different types of commercial incubators were selected in this study:No.1 large box incubators;No.2 small box incubators;No.3 desktop dry incubators;No.4 desktop wet incubators.The commercial thermometer detector was coupled to the cover of the Petri dish and a simple device was set up to monitor temperature in the micro-drops.The Petri dish was removed from the CO2incubator and placed on a hot plate,and the temperature of micro-drops in the Petri dish was measured every 30 seconds.When the temperature dropped to 30℃,the Petri dishes were returned to four different incubators,and the rewarming time was recorded when the temperature was restored to 37℃. Results:The temperature in the micro-drops gradually reduced with the time left after the Petri dishes were placed on the thermo plate.After 3 minutes,the temperature reduced to 36.50(±0.52)℃.After 5 minutes and 10 minutes,the temperature reduced to 36.17(±0.67)℃ and 35.53(±0.64)℃ respectively.After 10 min,the droplet temperature stabilized.When the Petri dishes were returned to the incubator,the time of the micro-drops temperature from 35℃ to 37℃ was taken 5.58(±1.37)min,15.69(± 5.19)min,5.21(± 0.45)min,4.88(±0.51)min respectively in four different incubators.The rewarming time for No.2 incubator was significantly longer than other incubator(P<0.05).And the rewarming time for No.4 incubator was shortest,and there were no significant difference among the other incubators(P>0.05). Conclusions: From the temperature point of view,it is suggested that the time of manipulating embryos on the thermo plate should be controlled within 3 min.And the time to return the optimal temperature was different when the Petri dishes are put back to different incubators.The incubator with shortest rewarming time should be chosen to optimize the effect of embryo culture.关键词
