纤维素降解菌的筛选、酶活及对稻草秸秆的降解研究
Study on Screening Cellulose-degrading Fungi and its Enzyme Activity and the Effect on the Degradation of Rice Straw

Abstract
The strains were cultured in the medium with filter strips as the sole carbon source and isolated with PDA medium from soil samples with rich cellulose in Hengyang. Then cellulose-degrading fungi were selected in Congo red cellulose agar. One strain (numbered 1-4) was screened by the ratio colony diameter to diameter of transparent circle and colony diameter. The strain was identified by the morphological characteristics on the identification culture medium (CYA) with three inoculation method and inserted cover glass. Then the strain was inoculated to liquid fermentation cultivation for 5 to 9 days and took its centrifugal filtrate as crude enzyme liquid to test its cellulose-degradation ability by filter paper enzyme activity. Last the strain was inoculated in pretreated rice straw solid culture medium to explore its straw-degrading capability. The strain was identified as Penicillium donkii. FPAase of the fungus reached the maximum (15.0 U/mL) at the 8th day under the condition of 28℃, 180 r/min. The straw solid medium were inoculated with the strain for 21 d, the cellulose content was reduced from 30.30% to 26.36%. The strain was identified P. donkii. It has some degradation ability to cellulose and can quicken the tempo of rice straw degradation.关键词
