Influence of cytochrome P (CYP) 1A1 gene polymorphism on the clinical outcomes of docetaxel plus capecitabine in the treatment of metastatic breast cancer

Abstract
Objective To investigate the association between cytochrome P (CYP) 1A1 polymorphism and clinical outcomes in metastatic breast cancer (MBC) patients treated by docetaxel plus capecitabine.Methods Sixty-nine MBC patients who were treated with docetaxel plus capecitabine were included in this study.And 79 single nucleotide poly morphisms (SNPs) in CYP450,whose minor allele frequency (MAF) ≥ 10% were genotyped.Association between rs1048943 (Ile462Val) and clinical outcomes were analyzed.Results Only one SNP,CYP1A1 rs1048943 was sigrnificantly associated with progress free survival (PFS),the median PFS of GA/GG genotype patients and wild type AA genotype patients were 8.3 months (95 % CI:5.4-11.8) and 5.3 months (95% CI:2.9-7.6),with significant difference (P <0.05).Multivariate analysis conrirmed its prognostic significance for PFS (HR =0.4,P < 0.05).Conclusion CYP1A1 rs1048943 polymorphism is probably a potential prognostic marker for survival outcomes treated with docetaxel plus capecitabine chemotherapy in MBC patients.关键词
