Abstract

Avian influenza virus poses a great threat to not only poultry, but also human beings. Several human pandemics resulted from the reassortant of avian and human influenza viruses. Four reassortant viruses, including r2093HANAPB2/WSN, r2093HANAPB1/WSN, r2093HANAPA/WSN and r2093HANANP/WSN, were rescued by reverse genetics, each of which contains the HA gene, NA gene, and one of polymerase genes or NP gene of A/Chicken/Shanghai/2093/2009, and other 5 genes of A/WSN/1933(H1N1). The replication capability of the reassortant viruses were tested on MDCK, DF1 and A549 cells in mice. The results showed that virus titers of all these reassortant viruses reached the peak on MDCK, DF1 and A549 cells infected with 0.001 multiplicity of infection (MOI) at 24 hours post infection, while r2093HANAPB1/WSN and r2093HANAPA/WSN replicated better than r2093HANAPB2/WSN and r2093HANANP/WSN in those cells. Accordingly, r2093HANAPB1/WSN and r2093HANAPA/WSN showed higher replication efficiency than r2093HANAPB2/WSN and r2093HANANP/WSN in nasal turbinates of mice inoculated intranasally, although the titers of those viruses did not show clear difference in the mouse lungs. It is suggested that the replication capabilities of the reassortants with different polymerase genes or NP gene are significantly different and the molecular mechanism causing the difference is worth to be explored further.
