甲状旁腺素联合唑来膦酸对骨质疏松骨折愈合的作用
Effects of parathyroid hormone combined with zoledronic acid on healing of osteoporotic fracture

Abstract
Objective To study the effects of parathyroid hormone ( PTH) combined with zoledronic acid (ZA) on fracture healing in ovariectomized mice. Methods Ninety six 12-week C57BL/6 female mice were enrolled in this study. These mice received transverse fracture at the lower third of left femur after being ovariectomized one month lat-er. The mice were then randomly divided into four groups and treated with normal saline, PTH1-34, zoledronic acid (ZA), and PTH1-34 plus ZA. Eight mice were killed at the second and fourth week respectively in each group. Micro-CT, biomechanical test, histology and RT-PCR were performed. Results At the 2nd week after administration, bone mineral density (BMD) [ (0. 062±0. 009) g/cm3], bone volume (BV) [ (34. 8±2. 6) mm3], relative bone volume (BV/TV)[ (52. 3±3. 0)%] and trabecular bone thickness (Tb. Th) [ (0. 212±0. 031) μm] in PTH1-34 plus ZA group were significantly higher than saline group (P<0. 05). At the 4th week after administration, BMD[ (0. 072± 0. 010) g/cm3] and BV/TV[ (60. 3±5. 2)%] were significantly higher than other groups (P<0. 05). Whether at the 2nd or 4th week after administration, bending ultimate force [2nd and 4th week: (6. 1±0. 7) N, (10. 8±1. 6) N] and bending rigidity [2nd and 4th week: (11. 6±1. 1) N/m, (22. 1±2. 8) N/m] were significantly higher than other groups (P<0. 05). Histologically, at the 2nd week after administration, the trabecular bone in PTH1-34 plus ZA group had the highest density, and at 4th week, the marrow cavity showed good recanalization. At the 2nd week after administra-tion, the expression of ALP in PTH1-34 plus ZA group was significantly higher than that of other groups (P<0. 05). At the 4th week after administration, the expression of BSP in PTH1-34 plus ZA group was higher than that in other groups (P<0. 05). Conclusion PTH combined with ZA has a superimposed effects on fracture healing in ovariectomized mice.关键词
