Abstract

To study the chemical constituents from the sporophore of Ganoderma luteomarginatum and their cytotoxic activities. Methods: The compounds were separated and repeatedly purified by column chromatography over silica gel and Sephadex LH-20,preparative TLC, and semi-preparative HPLC. Their structures were elucidated by NMR and MS spectroscopic methods. Furthermore, the cytotoxicities of the chemical components for mouse colorectal carcinoma CT26 cell line were measured by MTT method. Results: Seven ergostane-type steroids were isolated from Ganoderma luteomarginatum, and their structures were identified as 5α,8α-epidioxyergosta-6,22-dien-3β-ol (1),5α,8α-epidioxyergosta-6,9 (11),22-triene-3β-ol (2),ergosterol (3),ergosta-7,22-dien-3β-ol(4), ergosta-7, 22-dien-3-one(5), calvasterol A(6) and volemolide(7). Compounds 1~ 5 inhibited the growth of CT26 lines with IC50 values of 76. 93, 37. 19, 13. 02, 48. 19, and 1. 25 μmol/L, respectively. Conclusion: All compounds are isolated from Ganoderma luteomarginatum for the first time, compounds 6 and 7 are first reported from the Ganoderma genus, and compounds 1~ 5 show significant cytotoxic activities against mouse colorectal carcinoma CT26 cell line.
