摘要

Abstract

Objective: To explore the effect of Herba Siegesbeckiae on oxidative stress of synociocytes induced by Monoso-dium Urate (MSU) mediated by Nrf2/HO - 1 signal pathway. Methods: The knee synovial tissues of the rats were taken and synoviocytes were separated and cultured in vitro. The synoviocytes were medicated by different serum containing Herba Siegesbeckiae, the content of ROS, SOD and MDA were measured by ELLSA method. The expressions of Nrf2 and HO -1 were detected by RT - PCR technique. Results: ELISA assay showed that compared to the control group, the contents of ROS and MDA of the synovial cells increased significantly, whereas the content of SOD decreased significantly in the model group. ELISA analysis also confirmed the content of ROS and MDA reduced and the content of SOD increased, in the Herba Siegesbeckiae groups of different concentrations compared to the model group. In terms of the mR-NA expressions of Nrf2 and HO - 1 in synovial tissue, they were increased in the Herba Siegesbeckiae group. Conclusion:Herba Siegesbeckiae can up - regulate Nrf2 synthesis and nuclear translocation through regulating synovial cells of Nrf2/HO - 1 signal pathway, thus to promote HO - 1 expression and inhibit the reaction of oxidative stress.
