摘要

Abstract

Objective: To observe the changes of urine metabolites in spontaneously hypertensive rats( SHRs) before and after the intervention of Coptis chinensis Franch., to find the potential biomarker of hypertension, and to explore the anti-hypertensive mechanism of Coptis chinensis Franch.. Methods: The SHRs were divided into Valsartan group, Coptis chinensis Franch. group and the model group, and the Wistar-Kyoto rats were taken as the normal control. Urine sam-ples were separated and identified by HPLC/TOF-MS in positive polarity. The obtained data was normalized and ana-lyzed by using univariate and multivariate statistical analyses, further partial least-square discriminant analysis( PLS-DA) and variable importance factors were processed to see the cluster among groups. Results: Coptis chinensis Franch. can lower mean arterial pressure of SHRs. Finally, eleven identified biomarkers were considered to be involved in trypto-phan metabolism(indoleacetic acid, 5 -Hydroxyindoleacetic acid, quinaldic acid), pyrimidine metabolism(cytidine, L-dihydroorotic acid), arachidonic acid metabolism(prostaglandin E2, 11-dehydro-thromboxane B2) and primary bile acid biosynthesis( glycocholic acid). Conclusion: Coptis chinensis Franch. can make the metabolisms tendency of these biomarkers mormal in SHRs, and its antihypertensive mechanism may be related with the inflammation response and lipid regulation.
