芪连扶正胶囊编辑肺癌干细胞免疫重塑的研究
Immune Remodeling of LCSCs Intervened by Qilian Fuzheng Capsule
摘要
Abstract
Objective: To observe the effect Qilian Fuzheng Capsule on the expressions of immune related factors in lung cancer stem cells( LCSCs) and explore the related mechanism. Methods: Lung cancer A549 cells were cultured in vitro, LCSCs were sorted out by immunomagnetic beads, finally serum containing of Qilian Fuzheng Capsule was prepared. LCSCs were intervened according to experimental grouping; afterwards, the expressions of IL-10 and TGF-β1 in each group were detected by Elisa method; the expression of Foxp3 was detected by Western blot method, and TGF-β1 mR-NA expression was detected by RT-PCR technique. Results: In terms of expressions of IL-10, TGF-β1, Foxp3, and TGF-β1mRNA, they were significantly decreaed in all medication groups(Qilian Fuzheng Capsule group, Cisplati-num group, Qilian Fuzheng Capsule +Cisplatinum group) compared to those in the control group ( P <0.05, P<0.01); of which, the differences of the group of Qilian Fuzheng Capsule plus Cisplatin were most obvious, it showed that the combination of Qilian Fuzheng Capsule and Cisplatin had synergistic effect. Conclusion: Qilian Fuzheng Capsule can reduce the expressions of IL-10, TGF-β1 and Foxp3 in LCSCs, it can improve the cellular immunosuppression and regulate immune remodeling to inhibit lung cancer metastasis.关键词
