Inhibitory Effect of Astragalus Polysaccharides on the Growth of Human Gastric Cancer Cell Line MKN45 and MGC-803

Abstract
Objective: To study the effect of astragalus polysaccharides (APS) on the growth of human gastric cancer cell line MKN45 and MGC-803. Methods: Human gastric cancer cells MKN45 and MGC-803 were placed into culture medium RPMI1640 for subculturing. Cells were divided into control group,5-fluorouracil (5-FU) group and APS group. The control group received 1640 culture medium for culturing; 5-FU group received different concentrations of 5FU for culturing; APS group received different concentrations of APS for culturing. Methyl thiazolyl tetrazolium (MTT) experiment was adopted to test the inhibition of cell growth. Flow cytometry was used to test cell cycle. Fluorescence microscope was used to observe apoptosis. Results: ①After culturing for 24 h,with APS concentration ≥10.0 g·L - 1 and 5-FU concentration ≥0.05 g·L - 1,MKN45 and MGC-803 cells were obviously inhibited and the difference was of statistical significance (P < 0.05); after culturing for 48 h,with APS concentration ≥ 5.0 g·L - 1 and 5-FU concentration ≥0.025 g·L - 1,MKN45 and MGC-803 cells were obviously inhibited and the difference was of statistical significance (P < 0.05). ②After culturing for 48 h,MKN45 and MGC-803 cell cycles of 5-FU group and APS group were all changed, the proportion of cells staying at G0 /G1 phase was obviously higher than that of control group, cells at S phase and G2 /M phase were obviously fewer than those of control group, and the differences were of statistical significance (P < 0.05). ③After culturing for 48 h, apoptosis rates of MKN45 and MGC-803 cells of 5-FU group and APS group were higher than that of control group and the difference was of statistical significance (P < 0.05). Conclusion: APS can inhibit the growth of gastric cancer cells MKN45 and MGC-803 and its possible mechanism of action is to make the cells stay at G0 /G1 phase and induce apoptosis.关键词
