Effect of Si Shi Chang Fu Prescription on Immune Function and Antioxidant Capacity of Cadmium Infected Rats

摘要

Abstract

Objective: To study the effect of Si Shi Chang Fu prescription on the interleukin-2(IL-2) and superoxidase dismutase(SOD) activity in rats induced by exposure to cadmium, to explore its mechanism. Methods: 60 SPF Wistar rats were randomly divided into blank group,model group,acetylcysteine group,Si Shi Chang Fu prescription low dose group,middle dose group and high dose group. The rats were killed 24 hours after the last administration, and the thoracic spleen index and spleen index were calculated. The activities of superoxide dismutase (SOD) and malondialdehyde (MDA) level in serum of rats were measured by colorimetric analysis. The serum levels of interleukin-2 (IL-2) and transforming growth factor- β1(TGF- β1) were measured by ELISA. Results: Weight in the blank group,model group, acetylcysteine group,Si Shi Chang Fu prescription low dose group,middle dose group and high dose group were (193.13±37.48) g,(144.40±11.55) g,(185.10±25.89) g,(162.88±13.11) g,(165.77±15.62) g,(179.20±22.73) g, respectively; indexes of spleen and thymus levels in above groups were (3.96±0.70 /1.69±0.50) mg·g - 1,(1.86±0.43 /0.72±0.33) mg·g - 1,(2.80±0.61 /1.33±0.25) mg·g - 1,(2.49±0.36 /0.83±0.22) mg·g - 1,(2.90±0.53 / 0.87±0.23) mg·g - 1,(3.03±0.72 /1.04±0.39) mg·g - 1, respectively; SOD level in above groups were (72.72±8.10) U·mL - 1,(38.28±4.92) U·mL - 1,(64.87±10.15) U·mL - 1,(56.70±8.94) U·mL - 1,(63.27±7.15) U·mL - 1,(65.54±4.92) U·mL - 1, respectively; MDA level in above groups were(6.26±0.98) nmol·L - 1,(13.36±1.15) nmol·L - 1,(7.86±0.83) nmol·L - 1,(8.09±0.98) nmol·L - 1,(7.19±1.24) nmol·L - 1,(7.00±0.85) nmol·L - 1, respectively; IL-2 level in above groups were (223.61±28.59) ng·L - 1,(130.56±20.18) ng·L - 1,(201.39±23.81) ng·L - 1,(176.67±20.75) ng·L - 1,(187.50±30.16) ng·L - 1,(193.33±38.37) ng ·L - 1, respectively;TGF-β1 level in above groups were (5.50±0.51) μg·L - 1,(11.78±0.83) μg·L - 1,(6.17±0.35) μg·L - 1,(7.93±0.60) μg·L - 1,(7.22±0.78) μg·L - 1,(6.40±0.43) μg·L - 1, respectively. Compared with the blank group, the wight of the model group was significantly reduced (P < 0.05); and the wight of each intervention group was higher than that of the model group (P < 0.05). Compared with the blank group, the spleen and thymus indexes of the model group decreased; and the spleen index increased in each intervention group than that in model group and the thymus index increased in acetylocystcine group than that in model group (P < 0.05). Compared with the blank group, the activity of MDA in the model group was significantly increased(P < 0.05). The level of MDA in each Si Shi Chang Fu prescription intervention group was significantly lower than that in model group (P < 0.05). Compared with the blank group, the level of IL-2 in the model group was significantly lower than that in the blank group, and the level of TGF-β1 was significantly higher than that of blank group(P < 0.05), and the level of IL-2 in each Si Shi Chang Fu prescription intervention group was significantly higher than that in the model group,TGF-β1 was significantly lower than that in the model group(P < 0.05). Conclusion: Si Shi Chang Fu prescriptions has protective effect on oxidative damage and immunity function damage induced by cadmium in rats, its mechanism is achieved by increasing the levels of SOD and IL-2.
