摘要

Abstract

Objective: To establish an HPLC-DAD method for simultaneous determination of 6 types of chlorogenic acids in Artemisia myriantha. Methods: The separation was achieved on a Kromasil 100-5 C18 analytical column (150 mm×4.6 mm, 5 μm) with a gradient elution of 0.1% acetic acid (A) and acetonitrile (B) at a flow rate of 1.0 mL·min-1. The detection wavelength was 327 nm and the column temperature was 25 ℃. Results: Perfect chromatogram with good separation of 6 phenolic acids including neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, 4,5-dicaffeoylquinic acid, 3,4-dicaffeoylquinic acid and 3,5-dicaffeoylquinic acid was obtained within 50 minutes. All the calibration curves showed good linearity between concentration of each compound and peak area. And the values of r were no less than 0.999,7 for all the analytes. The recoveries were 98.06%-98.96%. The developed method was successfully applied for the analysis of 6 chlorogenic acids in Artemisia myriantha obtained from 6 different provinces. Conclusion: The developed method is accurate and reliable, and can be helpful for determination of chlorogenic acids and quality control of Artemisia myriantha.
