Abstract
Objective: To establish a method for determination of alanine in sparrow brains by pre-column derivatization and reversed-phase high performance liquid chromatography and compare samples in different collection times. Methods: The chromatographic column was the Elite C18 (200 mm×4.6 mm, 5 μm). The mobile phase A was acetonitrile-water (1∶1) and the mobile phase B was 0.05 mol/L, with sodium acetate buffer (pH= 6.4) and gradient elution. The detection wavelength was 360 nm, and the flow rate was 1.0 mL/min. Results: The linear relation of alanine content determination was good in 0.04-0.2 μg, with average recovery rate of 99.56％ and RSD％ of 1.67％. The content of alanine in sparrow brains of different batches was 3-5 mg/g. Conclusion: The method for determination of alanine in sparrow brains by pre-column derivatization and reversedphase high performance liquid chromatography shows good repeatability and high sensitivity and it can be used for quality control of sparrow brains.关键词
