摘要

Abstract

Peanut(Arachis hypogaea) is an important crop,but it is badly affected by drought stress.The draft peanut genome sequence released in 2016 can be used to help understand the molecular mechanisms of drought tolerance in this species.Transcription factors (TFs) play central roles in defense responses in peanut.In this study,the differential expression of TFs in response to drought stress in the drought-resistant peanut cultivar Jll by RNA-seq was analyzed.Pooled samples obtained at 6,12,18,24,and 48 h were compared with control samples at 0 h.In total,this study identified 236 differentially expressed TFs based on the transcriptome data;142 were up-regulated and 94 were down-regulated.Among the differentially expressed TFs,members of the MYB and bHLH families were the most abundant.In up-regulated TFs group,the fold change range was from 2.034 1 to 43.968 3,and 69 TFs was>10.In down-regulated TFs group,the fold change range was from-2.1031 to-11.6103,and 58 TFs was <-4.In this study,25 MYB genes were induced and 17 were down-regulated;17 bHLH genes were up-regulated and 14 were down-regulated;all NAC TFs was used to were up-regulated.In order to validate the gene expression data obtained by RNA-seq,qRT-PCR was used determine the expression levels of 14TF genes.Although the data obtained from qRT-PCR and RNA-seq were different,the trends in the expression levels determined by qRT-PCR were consistent with those obtained by RNA-Seq,thereby confirming the accuracy of the RNA-seq results.The results confirm the idea that drought resistance in peanut was largely dependent upon multiple TF families.The results of this study will provide a useful resource for further molecular research in this species.
