口疮病毒ORFV125蛋白抑制宿主细胞凋亡的研究
Study on host cell apoptosis inhibited by ORFV125 protein

Abstract
Previous study showed that ORFV125 protein and Bcl-2 protein family had highly similar 3Dstructure and domain. To explore whether ORFV125 protein also had anti-apoptotic function as well as Bcl-2, the domain mutation including BH1, BH2, BH3 and BH4, and the deletion mutation BH1, BH2, BH3 and BH4 were constructed, respectively. Apoptosis levels were detected by flow cytometry after transfection with these domain mutations in 293Tcells for 24 h and 72 h, respectively. In result, the percents of early apoptosis were lower than 5%, and the percents of late apoptosis were no more than 6% at 24 hour;the percents of early apoptosis were higher than 20%, and the percents of late apoptosis were no more than 4% at 72 hour. There were no significant differences between different mutants at the same transfection time. These results indicated that ORFV125 protein and its mutants had anti-apoptosis function, which provided theoretical basis for further study on ORFV125 protein.关键词
