Abstract

One strain of fowl adenovirus serotype 4 named HZ-06-01(FAdV-4)was isolated from Guangdong Province in 2016. To investigate the biological characteristics of this strain, the plaque purification was used to purify the isolated virus on LMH cells. The isolated viral sequences of hexon, ORF19 and ORF27 genes were analyzed, revealing that the identity was 100% between HZ-06-01 and Chinese representative strains HB1510 and JSJ13, and it existed ORF19 and ORF27 gene deletion. Two groups of SPF chickens(three weeks age-old) were challenged by intramuscular and oral administration, respectively. The chickens of intramuscular infection group had higher mortality rate 100%(10/10), as well as broadly viral detection in multiple organs in dead chickens. However, the chickens of oral administration group also were induced specific antibody on day 14 post-infection and the mortality was 0%(0/10). Histopathological examination showed that the liver had obvious nucleus inclusion bodies, severe reduction and necrosis of lymphocytes showed in spleen at dead chickens. In conclusion, HZ-06-01 belonged to FAdV-4, and SPF chickens showed obvious HHSby intramuscular administration. The results of this study provide the basic data for the diagnosis and prevention of the disease.
